چکیده
Research question: Do miRNAs play a role in regulating endoplasmic Reticulum Stress (RS) and Unfolded Protein Response (UPR) decidualized cells endometrium associated with reproductive failures?Design: Endometrial stromal cell line St-T1b was vitro 8-Br-cAMP during 5 days, or treated the RS-inducer Thapsigargin (Tg). Expression of RS-sensors, UPR markers potential miRNA regulators were analysed by qPCR. biopsies from patients recurrent pregnancy loss (RPL) implantation failures (RIF) investigated for location expression. Results: After decidualization, increased expression RS-sensors including ATF6α, PERK, IRE1α, marker, CHOP. TXNIP, which serves as link between RS-pathway inflammation, well inflammasome NLRP3 IL-1β cells. An silico analysis identified miR-17-5p, miR-21-5p, miR-193b-3p potentially involved regulation RS/UPR pathways inflammation embryo implantation. Their significantly decreased non-decidualized presence Tg. Finally, selected localized situ hybridization glandular epithelial endometrial samples RPL RIF patients. stroma lower comparison those patients. Conclusions: Decidualization is accompanied processes an inflammatory response that influenced miR-193b-3p. These are differentially expressed indicating alteration pathways.

Perturbation of endoplasmic reticulum (ER) homeostasis results in a stress condition termed "ER stress" determining the activation of a finely regulated program defined as unfolded protein response (UPR) and whose primary aim is to restore this organelle's physiological activity. Several physiological and pathological stimuli deregulate normal ER activity causing UPR activation, such as hypoxia...
متن کاملEndoplasmic reticulum stress: signaling the unfolded protein response.
The endoplasmic reticulum (ER) is the cellular site of newly synthesized secretory and membrane proteins. Such proteins must be properly folded and posttranslationally modified before exit from the organelle. Proper protein folding and modification requires molecular chaperone proteins as well as an ER environment conducive for these reactions. When ER lumenal conditions are altered or chaperon...
متن کاملEndoplasmic reticulum stress sensing in the unfolded protein response.
Secretory and transmembrane proteins enter the endoplasmic reticulum (ER) as unfolded proteins and exit as either folded proteins in transit to their target organelles or as misfolded proteins targeted for degradation. The unfolded protein response (UPR) maintains the protein-folding homeostasis within the ER, ensuring that the protein-folding capacity of the ER meets the load of client protein...
متن کاملAutophagy Counterbalances Endoplasmic Reticulum Expansion during the Unfolded Protein Response
The protein folding capacity of the endoplasmic reticulum (ER) is regulated by the unfolded protein response (UPR). The UPR senses unfolded proteins in the ER lumen and transmits that information to the cell nucleus, where it drives a transcriptional program that is tailored to re-establish homeostasis. Using thin section electron microscopy, we found that yeast cells expand their ER volume at ...
متن کاملThe PERK Arm of the Unfolded Protein Response Regulates Mitochondrial Morphology during Acute Endoplasmic Reticulum Stress
Endoplasmic reticulum (ER) stress is transmitted to mitochondria and is associated with pathologic mitochondrial dysfunction in diverse diseases. The PERK arm of the unfolded protein response (UPR) protects mitochondria during ER stress through the transcriptional and translational remodeling of mitochondrial molecular quality control pathways. Here, we show that ER stress also induces dynamic ...
